(E/Z)-BIX02189 is a selective inhibitor of MEK5 and ERK5 that shows no inhibition against closely related kinases MEK1, MEK2, ERK2, and JNK2. It selectively blocks sorbitol-stimulated phosphorylation of ERK5 without affecting ERK1/2 phosphorylation in HeLa cells. Downstream, it suppresses the transcriptional activity of MEF2C, disrupting downstream functions of the MEK5/ERK5 signaling cascade. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
