Avasimibe (CI-1011) is an orally bioavailable acyl-CoA:cholesterol acyltransferase (ACAT) inhibitor that also targets cytochrome P450 isoenzymes CYP2C9, CYP1A2, and CYP2C19. By inhibiting ACAT, it blocks intracellular cholesterol esterification, leading to reduced hepatic secretion and pool sizes of VLDL and LDL apolipoprotein B. Additionally, Avasimibe induces CYP3A4 and multidrug resistance protein 1 expression, altering lipid homeostasis and drug metabolism pathways. [1]
