Biological Activity

Amprenavir (VX-478) is a potent HIV protease inhibitor (Ki 0.6 nM) and a selective PXR agonist. By binding directly to the active site of HIV protease, amprenavir interrupts the maturation phase of the viral replicative cycle by blocking the cleavage of viral polyproteins. This prevents the formation of infectious viral particles and suppresses viral replication in HIV-infected cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
