Amlodipine besylate is a long-acting calcium channel blocker that inhibits calcium-induced contractions in depolarized rat aorta with an IC50 of 1.9 nM. It exhibits a slow onset of action compared to other dihydropyridine calcium antagonists and suppresses vascular smooth muscle contraction. By blocking L-type calcium channels, amlodipine decreases intracellular calcium influx, leading to peripheral vasodilation and reduced vascular resistance. [1]

- Preheat media/saline to 37°C.
