Biological Activity

Aloxistatin (E-64d) is a cell-permeable, irreversible inhibitor of cysteine proteases, including cathepsins B and L and calpains. By inhibiting intracellular cysteine proteases, aloxistatin blocks downstream proteolytic degradation pathways, impairs blood platelet aggregation, and suppresses viral processing. Consequently, aloxistatin markedly reduces cathepsin-dependent cellular entry of SARS-CoV-2 pseudovirions and restricts cathepsin-mediated protein cleavage in preclinical models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
