Biological Activity

Agomelatine (S20098) is a potent melatonin receptor agonist and 5-HT2C receptor antagonist, functioning as a norepinephrine-dopamine disinhibitor. By selectively antagonizing 5-HT2C receptors, agomelatine disinhibits and enhances norepinephrine and dopamine release in the frontal cortex. Simultaneously, activation of MT1 and MT2 melatonergic receptors resynchronizes circadian rhythms and promotes neurogenesis and neuroplasticity in the hippocampus, modulating downstream frontocortical dopaminergic and adrenergic pathways to exert its neurobiological functions. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
