Biological Activity

5-Ethynyl-2'-deoxyuridine (EdU) is a thymidine analogue that acts as a probe for measuring cellular DNA synthesis and cell proliferation. Upon incorporation into newly synthesized DNA during S-phase, EdU reacts with fluorescent azides via copper-catalyzed click chemistry ([3+2] cycloaddition), enabling rapid and sensitive detection of replicating cells without requiring DNA denaturation. Additionally, EdU serves as an alkyl chain-based linker in the synthesis of proteolysis targeting chimeras (PROTACs). [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
