Biological Activity

ZEN-3694 is an orally bioavailable bromodomain and extra-terminal (BET) protein inhibitor. By targeting BET epigenetic regulators, ZEN-3694 downregulates key cell cycle components, including CDK6 and CCND1, thereby reversing resistance to CDK4/6 inhibitors. Downstream, treatment with ZEN-3694 suppresses cellular proliferation and induces apoptosis in estrogen receptor-positive breast cancer cell lines by inhibiting cell cycle and cellular growth pathways. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
