Biological Activity
Zavondemstat is a small-molecule pan-inhibitor of histone lysine demethylase 4 (KDM4) that selectively targets KDM4A–D isoforms with no effect on other KDM families. By blocking KDM4-mediated histone demethylation, it regulates chromatin structure and downstream gene expression profiles. This epigenetic inhibition suppresses cell proliferation across diverse cancer cell lines and organoid models, reduces tumor-initiating cell populations, and exerts potent antineoplastic activity in xenograft models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
