Biological Activity
Zaprinast (M&B 22948) is a selective inhibitor of cGMP-hydrolyzing phosphodiesterase isoforms, primarily targeting PDE5 and PDE6. By blocking phosphodiesterase-mediated degradation of cGMP, zaprinast elevates intracellular cGMP levels and activates downstream cGMP-dependent signaling pathways. In neurobiological studies, post-training administration of zaprinast impairs spatial memory retrieval and modulates hippocampal PDE5 expression in rats, demonstrating its role in regulating synaptic plasticity and cognitive functions.
