Z4P is a blood-brain barrier-penetrant small-molecule inhibitor targeting inositol-requiring enzyme 1 (IRE1). By inhibiting IRE1 endoribonuclease activity, Z4P suppresses the unfolded protein response pathway, downregulates downstream transcriptional networks, and blocks pro-survival endoplasmic reticulum stress signaling. This enzymatic inhibition impairs tumor cell adaptation to microenvironmental stress, suppressing cell proliferation and demonstrating potential for the adjuvant treatment of glioblastoma.
