YKL-5-124 is a potent, selective, and covalent CDK7 inhibitor that exhibits marked selectivity over CDK2, CDK9, and CDK12/13. By covalently targeting the CDK7 subunit of the TFIIH complex, it inhibits carboxyl-terminal domain phosphorylation of RNA polymerase II, thereby disrupting transcriptional initiation and promoter-proximal pausing. Consequently, CDK7 inhibition suppresses oncogenic gene expression and impairs cell cycle progression, leading to robust antiproliferative effects in tumor models. [1]
