YD54 is an orally bioavailable proteolysis targeting chimera (PROTAC) that selectively acts as a SMARCA2 degrader. Targeted depletion of SMARCA2 disrupts the SWI/SNF chromatin remodeling complex, blocking downstream transcriptional programs crucial for cancer cell survival. Through this mechanism, SMARCA2 degradation exhibits robust antitumor activity in SMARCA4-mutant non-small cell lung cancer xenograft models. [1]
