Biological Activity

Y16 is an inhibitor of G-protein-coupled Rho guanine nucleotide exchange factors (GEFs) that blocks the interaction between leukemia-associated RhoGEF (LARG) and Rho (Kd = 80 nM). By preventing LARG-mediated Rho activation, Y16 lowers intracellular RhoA-GTP levels and blocks downstream Rho signaling pathways. Consequently, this disruption leads to suppressed cancer cell proliferation, migration, and invasive capacity in preclinical breast cancer models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
