XYD049 is a cereblon-based molecular glue degrader targeting GSPT1. By recruiting the CUL4-RBX1-DDB1-CRBN E3 ubiquitin ligase complex to GSPT1, it promotes targeted ubiquitination and proteasomal degradation of the protein translation factor. This action disrupts protein synthesis and downregulates prostate cancer-associated oncogenes, including AR, AR-v7, PSA, and c-Myc, resulting in potent growth inhibition of 22Rv1 castration-resistant prostate cancer cells and marked in vivo antitumor efficacy. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
