XRK3F2 is a small-molecule inhibitor targeting the ZZ domain of p62 (SQSTM1). By blocking the p62-ZZ domain, XRK3F2 blunts multiple myeloma-induced suppression of the osteogenic transcription factor Runx2 in osteoblasts. This inhibition restores osteoblast differentiation and matrix mineralization, counteracting myeloma-induced bone destruction. Consequently, XRK3F2 promotes new bone formation and bone remodeling in the tumor microenvironment in vivo. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
