XMD8-92 is a potent dual inhibitor of big MAP kinase 1 (BMK1/ERK5) and bromodomain-containing proteins, including BRD4. By targeting BMK1/ERK5, XMD8-92 blocks BMK1 autophosphorylation and prevents the downstream phosphorylation and degradation of promyelocytic leukemia (PML) protein. This pathway inhibition leads to PML stabilization, cell cycle arrest, and suppression of tumor growth across preclinical cancer models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
