XL092 is an ATP-competitive multi-targeted receptor tyrosine kinase inhibitor targeting MET, VEGFR2, AXL, and MER. Inhibition of these signaling pathways suppresses receptor phosphorylation and blocks angiogenic and oncogenic survival mechanisms. Downstream cellular responses include repolarization of tumor-associated macrophages from M2 to M1 phenotypes, inhibition of efferocytosis, and decreased tumor microvasculature, ultimately driving potent tumor growth inhibition and enhanced immune cell infiltration. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
