XL019 is a potent and selective Janus kinase 2 (JAK2) inhibitor that exhibits marked selectivity over JAK1, JAK3, and TYK2. By targeting the ATP-binding pocket of JAK2, XL019 blocks JAK2 catalytic activity and inhibits downstream STAT phosphorylation, particularly STAT3 and STAT5. This disruption of the canonical JAK/STAT signaling pathway suppresses cellular proliferation and induces apoptosis in JAK2-dependent cell lines. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
