WYE-125132 (WYE-132) is a potent, ATP-competitive mTOR inhibitor with an IC50 of 0.19 nM, displaying over 5,000-fold selectivity for mTOR relative to PI3Ks. By targeting the catalytic domain of mTOR, it dual-inhibits both mTORC1 and mTORC2 complexes. This blockage suppresses downstream phosphorylation of S6K1, 4E-BP1, and Maf1, leading to impaired protein synthesis, altered bioenergetic metabolism, and decreased cell growth and survival in cancer models.
