Wu-5 is a ubiquitin-specific peptidase 10 (USP10) inhibitor that targets USP10 activity to modulate ubiquitin-dependent protein degradation pathways. By blocking USP10-mediated deubiquitination, it promotes the degradation of oncogenic proteins such as internal tandem duplication-mutated FLT3 (FLT3-ITD). This loss of protein stability disrupts downstream survival and proliferative signaling, resulting in enhanced cell death and synergistically augmenting crenolanib-induced apoptosis in acute myeloid leukemia cells.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
