Biological Activity

WHI-P258 is a synthetic dimethoxyquinazoline derivative that acts as a Janus kinase 3 (JAK3) inhibitor with a Ki value of 72 μM. By targeting JAK3, WHI-P258 suppresses the JAK/STAT signaling pathway, preventing the phosphorylation and activation of downstream STAT proteins. This inhibition blocks cytokine-mediated signal transduction, impairing lymphocyte activation, proliferation, and immune cell responses. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
