Biological Activity

VX-984 (M9831) is an orally active, potent, selective, and ATP-competitive inhibitor of DNA-dependent protein kinase (DNA-PK). By inhibiting DNA-PK, it suppresses non-homologous end joining repair, blocks DNA-PKcs autophosphorylation, and causes the accumulation of DNA double-strand breaks. Consequently, VX-984 enhances the cytotoxic and radiosensitizing effects of ionizing radiation in various cancer models, including non-small cell lung cancer.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
