VU0238429 is a selective positive allosteric modulator of muscarinic acetylcholine receptor subtype 5 (M5, with selectivity over M1 (EC50 > and M3 (EC50 >, showing no potentiating activity at M2 or M4 receptors. Selective activation of M5 potentiates agonist-induced cholinergic signaling and intracellular calcium mobilization, offering a useful chemical probe to investigate M5 receptor function in the central nervous system. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
