VU0071063 is a small-molecule activator of heterologously expressed Kir6.2/SUR1 channels with an EC50 of 7 μM. Activation of these ATP-sensitive potassium channels dose-dependently and reversibly hyperpolarizes the pancreatic β-cell membrane potential. Downstream, this membrane hyperpolarization inhibits glucose-stimulated calcium entry and blocks insulin secretion, thereby modulating pancreatic cell excitability and hormone release. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
