Biological Activity

VU 0364770 is a positive allosteric modulator of metabotropic glutamate receptor 4 (mGlu4) with an EC50 of 1.1 μM. By selectively potentiating mGlu4 activation without significant activity at other mGlu subtypes, it modulates glutamatergic neurotransmission in the central nervous system. Activation of mGlu4 downstream signaling regulates synaptic transmission and attenuates motor deficits, demonstrating neuroprotective and functional circuit modulation in preclinical models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
