Biological Activity

VT3989 is an orally bioavailable, selective inhibitor of TEAD auto-palmitoylation that blocks the interaction between TEAD transcription factors and yes-associated protein (YAP). By disrupting the Hippo signaling pathway, VT3989 suppresses YAP/TAZ-driven transcriptional activity and downstream oncogenic gene expression. This inhibition impairs cellular proliferation and demonstrates potent preclinical antitumor efficacy in models of NF2-mutant solid tumors and malignant mesothelioma. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
