Vonoprazan is an orally active, potassium-competitive acid blocker (P-CAB) that inhibits H⁺, K⁺-ATPase activity in gastric microsomes with an IC50 ofat pH 6.5. By competing with potassium ions for binding to gastric H⁺, K⁺-ATPase, it potently blocks the final step of gastric acid secretion from parietal cells. This inhibition suppresses gastric acid secretion and elevates intragastric pH, demonstrating robust antisecretory activity. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
