Vimseltinib (DCC-3014) is a dual tyrosine kinase inhibitor of c-FMS (IC50 <0.01 μM) and c-Kit (IC50 0.1-1 μM). As a switch-control inhibitor, it blocks CSF1R (c-FMS) autophosphorylation, downregulating downstream macrophage signaling pathways. This target inhibition depletes tumor-associated macrophages and osteoclasts, suppressing synovial cell proliferation, tumor growth, and bone degradation in preclinical models.[1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
