Vilanterol is a potent and selective agonist of β2-adrenoceptor (β2-AR, pEC50 10.37), displaying weaker activity at β1-AR (pEC50 6.98) and β3-AR (pEC50 7.36). Activation of β2-AR stimulates intracellular cAMP accumulation, driving signaling through protein kinase A downstream pathways. This pathway cascade induces rapid and sustained airway smooth muscle relaxation, suppressing bronchoconstriction in preclinical models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
