Biological Activity

Vafidemstat (ORY-2001) is a brain-penetrant dual inhibitor targeting lysine-specific histone demethylase 1 (LSD1, KDM1A) and monoamine oxidase B (MAO-B). Inhibition of LSD1 and MAO-B modulates histone demethylation and chromatin remodeling pathways, regulating epigenetic gene transcription in the brain. These downstream epigenetic modifications modulate neuroinflammatory signaling, helping rescue cognitive deficits and restore behavioral functions in preclinical models of neurodegenerative disorders. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
