Biological Activity

UZH2 is a potent and selective inhibitor of METTL3 with an IC50 value of 5 nM in a TR-FRET assay. By inhibiting the METTL3/METTL14 RNA methyltransferase complex, UZH2 demonstrates intracellular target engagement and significantly reduces N6-methyladenosine (m6A) levels in polyadenylated RNA, thereby impairing m6A-mediated epitranscriptomic signaling in acute myeloid leukemia and prostate cancer cell lines. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
