UNC0642 is a potent and selective inhibitor of the histone methyltransferases G9a (IC50 < 2.5 nM) and GLP (IC50 < 2.5 nM), displaying over 300-fold selectivity over non-target kinases, GPCRs, and ion channels. By inhibiting G9a and GLP enzymatic activity, UNC0642 selectively decreases intracellular dimethylated histone H3 lysine 9 (H3K9me2) levels and regulates downstream gene expression, resulting in suppressed cell proliferation and phenotypic alterations in preclinical cancer models. - Preheat media/saline to 37°C.
