UNC0379 is a selective, substrate-competitive inhibitor of N-lysine methyltransferase SETD8 (IC50 = 7.3 μM), demonstrating high selectivity over 15 other methyltransferases. SETD8 specifically catalyzes the monomethylation of histone H4 at lysine 20 (H4K20me1), playing a vital role in chromatin regulation, DNA replication, and cell cycle control. Treatment with UNC0379 suppresses SETD8 catalytic activity and reduces intracellular H4K20me1 levels, inhibiting SETD8-mediated cellular processes.
