Biological Activity

UNC2025 HCl is a potent, orally bioavailable dual inhibitor of MER (IC50 0.74 nM) and FLT3 (IC50 0.8 nM), exhibiting approximately 20-fold selectivity over Axl and Tyro3. By blocking MER and FLT3 kinase activity, UNC2025 attenuates downstream oncogenic signaling pathways including STAT, AKT, and ERK, leading to inhibited colony formation and impaired cell proliferation in leukemia models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
