TYRA-300 is a 3-pyridylindazole analog that acts as a potent, reversible, and selective inhibitor of FGFR3 kinase. By selectively targeting FGFR3, it inhibits receptor autophosphorylation and suppresses downstream MAPK/ERK and PI3K/AKT signaling cascades. This blockade attenuates oncogenic pathway activation, thereby suppressing cell proliferation and survival in cancer cell lines harboring activating FGFR3 genetic mutations or oncogenic fusions. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
