TRULI is a potent ATP-competitive inhibitor of large tumor suppressor kinases 1 and 2 (Lats1/2). By blocking Lats1 and Lats2 activity in the Hippo signaling pathway, TRULI prevents Yap phosphorylation, promoting Yap nuclear accumulation and activation of downstream transcriptional programs. Consequently, TRULI induces cell proliferation in postmitotic tissues and supports early-stage proliferative regeneration of sensory hair cell receptors in the inner ear. [1]
