Biological Activity

Tulmimetostat (CPI- is a potent, orally bioavailable inhibitor of enhancer of zeste homolog 2 (EZH2). By targeting the catalytic subunit of Polycomb repressive complex 2 (PRC2), tulmimetostat suppresses global histone H3 lysine 27 trimethylation (H3K27me3) and alters transcriptional programs. These molecular changes impair tumor growth and enhance chemosensitivity to agents such as cisplatin, showing marked activity in ARID1A-mutant preclinical cancer models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
