Trelagliptin (SYR-472) is a potent, orally active, and highly selective dipeptidyl peptidase-4 (DPP-4) inhibitor. By inhibiting DPP-4 enzymatic activity, it prevents the rapid degradation of endogenous incretin hormones, including glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). This sustained elevation of active incretins stimulates glucose-dependent insulin secretion from pancreatic beta cells and suppresses glucagon secretion, promoting enhanced glycemic control in vivo.
