Tradipitant (VLY-686, LY686017) is a selective second-generation antagonist of the neurokinin-1 (NK-1) receptor. By blocking substance P signaling through the NK-1 receptor, it inhibits downstream neurogenic inflammatory pathways. This receptor blockade modulates central and peripheral neural circuits, demonstrating therapeutic potential in models of chemotherapy-induced nausea, depression, social anxiety disorder, and alcohol dependence. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
