TR-14035 is a dual α4β integrin antagonist targeting α4β7 integrin (IC50 = 7 nM) and α4β1 integrin (IC50 = 87 nM). By blocking α4β integrin interaction with cell adhesion molecules such as MAdCAM-1 and VCAM-1, it inhibits leukocyte adhesion and infiltration into inflamed tissues. Downstream, TR-14035 attenuates inflammatory signaling, suppressing airway hyperresponsiveness and eosinophil recruitment in animal asthma models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
