Biological Activity

TPC2-A1-N is a potent, cell-permeable agonist of two-pore channel 2 (TPC2) that evokes robust TPC2-dependent calcium responses with an EC50 of 7.8 μM in cell-based calcium-imaging assays. Selective activation of TPC2 by TPC2-A1-N triggers intracellular calcium release from endolysosomal stores into the cytosol, thereby regulating endolysosomal membrane potential, vesicle trafficking, and downstream lysosomal functional phenotypes.
