Biological Activity

Toyocamycin is an adenosine analog that acts as an inhibitor of XBP1 mRNA splicing and a specific inhibitor of CDK9 (IC50 79 nM). Blockade of the IRE1α/XBP1 pathway impairs endoplasmic reticulum stress responses and promotes apoptosis in cancer cells, whereas CDK9 inhibition suppresses RNA polymerase II phosphorylation to reactivate epigenetically silenced tumor suppressor genes.[1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
