TNG348 is an orally bioavailable allosteric inhibitor of ubiquitin-specific protease 1 (USP1). By inhibiting USP1 deubiquitinating activity, TNG348 prevents the deubiquitination of PCNA and FANCD2, leading to the accumulation of ubiquitinated protein substrates. This disruption of the translesion synthesis pathway and DNA damage tolerance mechanisms leads to synthetic lethality and inhibits cell proliferation in BRCA1/2-mutated and homologous recombination-deficient tumor models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
