Biological Activity

TMRE (tetramethylrhodamine ethyl ester) is a cell-permeant, lipophilic cationic fluorogenic dye used to evaluate mitochondrial membrane potential (ΔΨm). Driven by the negative electrical potential generated across the inner mitochondrial membrane during oxidative phosphorylation, TMRE selectively accumulates within active mitochondria. Loss or dissipation of ΔΨm during mitochondrial dysfunction or early apoptosis reduces intracellular TMRE fluorescence, providing a readout for cellular viability and cytochrome c release.[1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
