Tinlorafenib (PF-07284890) is a potent, brain-penetrant inhibitor targeting BRAF V600E and V600K mutations. By selectively suppressing oncogenic BRAF kinase activity, it blocks downstream ERK phosphorylation within the MAPK signaling pathway. This pathway disruption inhibits tumor cell proliferation and drives robust tumor regression in both subcutaneous and intracranial BRAF V600E-mutant melanoma xenograft models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
