Biological Activity

Tinengotinib (TT-00420) is a multi-kinase inhibitor that strongly inhibits Aurora A and Aurora B, while also suppressing FGFR1/2/3, VEGFR1, JAK1/2, and CSF1R. By targeting mitotic progression and tumor microenvironment pathways, it inhibits cell proliferation and downregulates genes essential for growth in triple-negative breast cancer. Furthermore, tinengotinib upregulates CXCL10 and CXCL11 expression and reduces tumor-associated macrophage infiltration. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
