Tideglusib is a potent, irreversible, non-ATP-competitive GSK-3β inhibitor. By inhibiting GSK-3β activity, it blocks tau hyperphosphorylation and decreases amyloid plaque accumulation in neural tissues. Suppression of GSK-3β modulates Wnt/β-catenin and NF-κB signaling pathways, thereby exerting marked neuroprotective effects, suppressing neuroinflammation, and enhancing adult neural progenitor cell proliferation in animal models of neurodegenerative diseases.
