THZ1 2HCl is a covalent CDK7 inhibitor that targets a remote cysteine residue located outside of the canonical kinase domain. Covalent inhibition of CDK7 disrupts the cyclin-dependent kinase-activating kinase complex and impairs RNA polymerase II C-terminal domain phosphorylation. Downstream, THZ1 suppresses genome-wide transcription, preferentially repressing super-enhancer-associated oncogenes and inducing cell cycle arrest and apoptosis in cancer cells. [1]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
