Thiamet G is a potent and selective O-GlcNAcase (OGA) inhibitor with a Ki of 21 nM, displaying 37,000-fold selectivity over human lysosomal beta-hexosaminidase. By blocking OGA activity, Thiamet G increases cellular levels of protein O-GlcNAcylation and decreases hyperphosphorylation of tau protein in neuronal models. This inhibition helps modulate protein homeostasis and maintain cellular stability within central nervous system pathways. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
